The2023Drug Safety and Efficacy StudyPhase 1, US, (NCT01677513) was conducted in France from December 2003 to May 2004. It was the first study conducted to investigate the safety and efficacy of levitra at 20 mg, 10 mg, and 20 mg twice daily as monotherapy for erectile dysfunction.
In this Phase 1/2 open-label, randomized, placebo-controlled, crossover study of 20 mg, 20 mg twice daily for four to eight weeks in 40 men with erectile dysfunction, the primary efficacy endpoint was the percentage of patients who reported a satisfactory response to the drug, compared with placebo. The secondary efficacy endpoint was the percentage of patients who showed improvement on the response question in the presence of sexual arousal. In all, 120 patients were enrolled. Twenty-four patients were randomized: 12 patients were treated with 10 mg levitra and the other 14 patients were treated with 20 mg levitra. The mean age of the patients was 64.5 years. The majority of patients were older than 60 years, had a prior diagnosis of cardiovascular disease, and had normal erectile function at baseline. The most common underlying medical problem was hypertension, most commonly diabetes and smoking. Levitra was well tolerated in all patients, with no serious adverse events. The safety profile of levitra was assessed in patients who had a baseline clinical response to the study drug (20 mg vs. 10 mg) and a response to treatment with either 20 mg or 10 mg levitra.
